YieldReaction ConditionsOperation in experiment
With Kluyveromyces lactis beta-galactosidase; at 37℃; for 24h;Enzymatic reaction; 4gl-1 of allyl alpha-d-gal and allyl beta-d-gal was incubated with 1Uml-1 enzyme in a total volume of 625mul at 37C with shaking for 24h. A sample was removed at various times and the reaction stopped by heating for 5min at 95C. It was mixed with an internal standard (glycerin, 50mgml-1) and analyzed with an HPLC fitted with a refractive index detector (column: YMC-Pack Polyamine II, YMC, 250×4.6mm, S: 5mum, 12nm, mobile phase: acetonitrile and water, 85:15, flow 1mlmin-1, temperature: 24C).
