Ack1 (Activated Cdc42-associated kinase 1, TNK2) is a member of the non-receptor tyrosine kinase family, characterized by an SH3 domain, a CRIB domain, and a tyrosine kinase catalytic domain. It is widely expressed in various tissues, particularly in tumor cells. Ack1 interacts with multiple signaling molecules, participating in the regulation of cell survival, proliferation, migration, and downstream signaling pathways such as PI3K/Akt and MAPK. The activity and expression levels of Ack1 are strictly regulated under specific conditions, and mutations or overexpression of its gene are closely associated with tumor occurrence, development, and drug resistance.
