YieldReaction ConditionsOperation in experiment
A 100 mL oven-dried round-bottom flask with a magnetic stirbarwas purged with N2. Diisopropylamine (138 mg, 193 mL,1.37 mmol) was added via syringe and dissolved in distilled THF(5 mL). The contents were stirred, then placed on a dry ice-bath.While kept at 78 C, n-butyllithium (850 mL, 1.37 mmol, 1.6 M inhexane) was then added via syringe and the contents were stirredfor 20 min. A solution of <strong>[1198-97-6]4-phenylpyrrolidin-2-one</strong> 4 (200 mg,1.24 mmol) dissolved in distilled THF (10 mL) was added to themixture via syringe and left to stir for 45 min. After this time, (S)-Naproxen chloride 16 (617 mg, 2.48 mmol) was dissolved indistilled THF (10 mL) and added via syringe. The contents werelifted off the dry ice-bath and allowed to stir at room temperaturefor 2.5 h. Saturated aqueous NaHCO3 (30 mL) was used to quenchthe mixture, which was then washed with dichloromethane (3 x30 mL). The organic layers were combined, dried with MgSO4,filtered and concentrated under reduced pressure to afford thecrude diastereomers as a solid. The diastereomers were separatedusing column chromatography (Mobile phase: 10% EtOAc in petroleumbenzine, TLCs were checked in 20% EtOAc in petroleumbenzine) to afford both (R,S)-17 and (S,S)-17.
