YieldReaction ConditionsOperation in experiment
44.1% With sodium carbonate; XPhos; In 1,4-dioxane; water; at 80℃;Inert atmosphere; Compound 1-c (1.5 g, 6.09 mmol), compound 27-c (prepared according to the method disclosed in: WO 2012/ 032067 Al) (2 g, 6.09 mmol), anhydrous sodium carbonate (1.94 g, 18.27 mmol), dioxane/water (20 mL/20 mE) and Pd(dppf)C12CH2C12 (500 mg, 0.6 mmol) were placed into a reaction flask. The reaction mixture was heated under nitrogen atmosphere to 80C. overnight. The reaction mixture was concentrated under reduced pressure, and then partitioned between dichioromethane (50 mE) and water (50 mE). The organic layer was separated, dried over anhydrous sodium sulfate, and concentrated under reduced pressure. The residue was purified by silica gel column chromatography to obtain compound 27-b (1.05 g, 44.1%), as a light brown solid. ECMS (ESI): mlz=449 (M+H).
