YieldReaction ConditionsOperation in experiment
50% First, 4-(hydroxymethyl)phenylboronic acid pinacol ester (400 mg, 1.7 mmol) was added to dichloromethane (10 mL), followed by the addition of triphosgene (according to 15 wtpercent in toluene, 6 mL, 8.4). Mm)It was stirred at room temperature for 36 hours to complete clarification. After completion of the reaction, the solvent and the unreacted triphosgene were removed by rotary evaporation, and t-butyl-N-methyl-N-[2-methylamino-ethyl]carbamate (320 mg, 1.7) dissolved in dichloromethane was quickly added. Mm). Then 10 mL of dichloromethane and triethylamine (517 muL, 3.7 mmol) were added and stirred at room temperature for 10 minutes, and finally the reaction was stopped by adding 1 mL of methanol. The reaction mixture was concentrated and subjected to column chromatography (mobile phase: n-hexane: ethyl acetate = 4:1 to 2:1) to give a colorless oil of Boc-BE (391 mg, yield 50percent).
