A mixture of 4-nitrothiophenol (1.44 g), <strong>[178488-39-6]5-methylimidazo[1,2-a]pyridin-3-yl methanol</strong> (1.5 g), concentrated hydrochloric acid (15 ml) and acetic acid (15 ml) was stirred for 24 hours at 100C. The mixture was neutralized with 12N sodium hydroxide at 0C, and extracted with ethyl acetate. The organic layer was washed with saturated brine, dried over magnesium sulfate, and concentrated under reduced pressure. The obtained residue was separated and purified by column chromatography (ethanol:ethyl acetate 1:9), to give 5-methyl-3-[[(4-nitrophenylthio)methyl]imidazo[1,2-a]pyridine (2.03 g) as yellow crystals. mp 167 to 169C 1H-NMR (200 MHz, CDCl3) δ 3.00 (3H, s), 4.76 (2H, s), 6.59 (1H, d, J = 6.8 Hz), 7.12 (1H, dd, J = 9.0, 6.8 Hz), 7.38 (2H, d, J = 9.0 Hz), 7.47 to 7.52 (2H, m), 8.16 (2H, d, J = 9.0 Hz).
