93% With sodium hydride; In methanol; mineral oil; at 120℃; for 12h;Cooling with ice; Reference Example-69 4-Chloro-2,5-difluorobenzoic acid (25.0 g, 126 mmol) and methanol (5.6 mL, 139 mmol) were added to a suspension of a 55% oil dispersion (10.99 g, 252 mmol) of sodium hydride in DMF (250 mL) under ice-cooling, followed by stirring at 120 C. for 12 hours. After the reaction was completed, the reaction solution was cooled to room temperature, and concentrated hydrochloric acid was added until the reaction solution became acidic, and the resultant product was extracted with diethyl ether (100 mL*3). The combined organic layer was washed with water, dried over magnesium sulfate, concentrated under reduced pressure, and dried under reduced pressure, whereby 4-chloro-2-fluoro-5-methoxybenzoic acid (23.97 g, yield: 93%) was obtained as a white solid. 1H-NMR (400 MHz, CDCl3): delta4.07 (s, 3H), 7.11 (d, J=5.6 Hz, 1H), 7.93 (d, J=9.0 Hz, 1H). 19F-NMR (376 MHz, CDCl3): delta-122.8 (s, 1F).
