YieldReaction ConditionsOperation in experiment
98% With triethylamine; In tetrahydrofuran; at 50℃; for 5h; 30 mmol B-3 and 30 mmol A-4 were dissolved in THF 80 mL. Add 36 mmol of triethylamine, after heating to 50 C for 5 h, the reaction was quenched with water and the mixture was extracted twice with ethyl acetate. After recovering ethyl acetate, it was crystallized from ethyl acetate to give the product 10.1 g, yield 98%, HPLC purity ?99%.
11.3 g With D-Malic acid; N-ethyl-N,N-diisopropylamine; In ethanol; at 20℃; for 3h; <strong>[1218935-59-1](R)-2-(2,5-difluorophenyl)pyrrolidine</strong> (R)-2-hydroxysuccinate (17.6 g, 5.5 mmol, 1.1 eq) , 5-chloro-3-nitropyrazolo[1,5-a]pyrimidine (10.0 g, 5.0 mmol, 1.0 eq) and ethanol wasadded to a round bottom flask outfitted with a mechanical stirrer. After dissolution, DIPEA (22.7 g, 17.6 mmol, 3.5 eq) was added to the reaction mixture at room temperature. The reaction was stirred at room temperature for 3 hours. Filtered and the cake was washed with ethanol(30 mlx2). Dried at 40C to give the product (11.3 g).
